Tumor Necrosis Factor Receptor I (TNF RI)

Tumor Necrosis Factor Receptor I (TNFRI) is a 55 kDa pleiotropic cytokine that is considered a primary modifier of the inflammatory and immune reactions of animals produced in response to injury or infection. It is the receptor for TNFSF2; TNF-alpha and homotrimeric TNFSFI/lymphotoxin-alpha. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation and initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. It also contributes to the induction of non-cytocidal TNF effects including anti-viral state and activation of the acid sphingomyelinase.
